Effects of dietary supplementation with quercetin, δ-tocotrienol, or dexamethasone on the production of nitric oxide by thioglycolate-elicited peritoneal macrophages stimulated with LPS-plus IFN-β</ττ>. 4-week-old and 42-week-old C57BL/6 male mice were fed control diet (CD) or control diet supplemented with quercetin, δ-tocotrienol or dexamethasone for 4 week. After this, thioglycolate-elicited peritoneal macrophages were collected, adhered to the bottom of 12 well plates (1×107 cells/well in 1.0 mL medium) for 4 h, then washed with medium three times. Cells were treated with LPS (10 ng/well) or LPS (10 ng + IFN-β 50 U/well) for 18 h at room temperature, mixtures were centrifuged at 2000 rpm for 20 min. The supernatants were collected, and were assayed for production of NO by using Griess reagent [12]. Cell viability exceeded 95% in all experiments [13]. Data represent means ± SD; n = 3 mice per group, with triplicate analysis of each sample. Values in columns with different symbols differ at P< 0.05. Treatments 1 - 16 correspond to: 1 - 4 = 8-week-old mice or 5 - 8 = 46-week-old mice treated with 10 ng/well LPS; 9 - 12 = 8-week-old mice or 13 - 16 = 46-week-old mice treated with 10 n g LPS plus + 50 U/well IFN-β. CD = control diet; Quer = quercetin; Trie = δ-tocotrienol; Dexa = dexamethasone.
