Inhibition of NAMPT aggravates hepatic lipid accumulation in vitro. The hepatic steatosis was induced by oleic acid (OA, 0.5 mM) in HepG2 cells and the effects of FK866 (20 nM) on lipogenesis in hepatic cells were detected with or without NAD+ (1 mM) or NMN (100 μM) treatment for 24 h. a The images of Oil red O staining of HepG2 cells. b The intracellular TG level in HepG2 treated with FK866 or combined with NAD+ or NMN. c Quantitative analysis of the expressions of lipid synthetic genes by RT-PCR in HepG2 cells. d Western blots images (left) and quantitative analysis (right) of FASN expression in HepG2 cells treated with NAD+. e Western blots images (left) and quantitative analysis (right) of FASN expression in HepG2 cells treated with NMN. All results were analyzed based on three independent experiments. The data represent the mean ± SEM. *P < 0.05 and **P < 0.01 versus the control; # P < 0.05, # # P < 0.01 and # # # P < 0.001 versus FK866
