Fetuin-A levels in PBMCs from obese subjects with and without diabetes. a Total proteins were extracted from peripheral blood mononuclear cells (PBMCs) from obese subjects with and without diabetes (n = 12 each) and subjected to Western blotting. The blots shown here are representative of independent experiments with consistent results. Proteins extracted from HepG2 cells were used as a positive control and loaded on the gel at a level 10 times lower than that for PBMCs. Data are presented as fold change in non-diabetic obese subjects compared with diabetic obese subjects. b Total RNA was isolated from PBMCs and positive control HepG2 cells and subjected to qRT-PCR, as detailed in the Materials and Methods section. c Total proteins were extracted from cell lines and PBMCs and then subjected to Western blotting for fetuin-A. Human serum and plasma were used as a positive control.
