MonoMac 6 cells (4 × 106 cells/5 ml X-Vivo 15 on Nunclon 60 mm dishes) were treated with 1 or 10 μM arachidonic acid (AA), eicosapentaenoic acid (EPA) or docosapentaenoic acid (DPA) for 24 h. Then the cells were pelleted by centrifugation (400 × G, +4°C) and washed with ice-cold PBS. Total cellular RNA was isolated, reverse transcribed and the resulting cDNA was amplified as described in the methods. The MMP-9 expression was demonstrated by real-time PCR. Results represent the means ± SEM of four independent experiments. Only 10 μM AA increased the expression of MMP-9 mRNA statistically significantly.Back to article page
