Effect of 13-MTD on mitochondrial membrane integrity of SKBR-3 cells. Cells cultured for 24 h on coverlips were incubated with 0.25 mM fatty acid sample for 4 h, and loaded with MitoCapture (Biovision, USA) reagent for 20 min at 37°C under 5% CO2 atmosphere. Cells were observed with a fluorescence microscope using band-pass filter. MitoCapture aggregates and fluoresces red in the mitochondria of healthy cells. However, in apoptotic cells, MitoCapture cannot accumulate in the mitochondria, it remains as monomers in the cytoplasm, and fluoresces green. Counting of mitochondria disrupted cells in total number of 40 cells were made for 4 to 6 separate microscopic fields, and the percentages are shown (B). Data not sharing the same letter are significantly different at p < 0.05 by Tukey-Kramer test.
